Zusammenfassung
Im Gegensatz zu den raschen Fortschritten bei der Aufklärung genetischer und epigenetischer Ursachen von angeborenen Krankheiten entwickeln sich Therapieoptionen zögerlich. Seltene Erkrankungen werden oft spät richtig eingeordnet, Spontanverlauf, prognostische Parameter oder Kriterien für therapeutische Interventionen sind unbekannt. Die Patientenzahlen sind für eine systematische Evaluierung therapeutischer Ansätze gering; Wissenschaft und Industrie kümmern sich in erster Linie um häufige Krankheiten. Trotz Kenntnis der genetischen Ursache(n) ist die Krankheitsentstehung nur in wenigen Fällen so weit verstanden, dass sich kausale Therapieansätze ergeben. Dennoch gibt es eine Vielzahl von Eingriffsmöglichkeiten in die Kaskade der Kausalfaktoren, mit denen die Manifestation der Krankheit ganz oder teilweise verhindert, ihre Progredienz verlangsamt, Komplikationen verhütet werden können.
