Summary
Extracts from peas, soy beans, wheat and flax seed were separated by gel or ion-exchange chromotography. The carotene-bleaching-activity was tested in the eluates: The lipoxygenase isoenzymes with a pH optimum of 6,5 are very potent carotene oxidases if linoleic acid is present in the incubation medium. Such carotene oxidases occur in peas, soy beans and wheat. —In contrast the alcaline lipoxygenase isoenzyme from soy beans, the hydroperoxide isomerase from flax seed and the peroxydases from wheat and soy beans are very poor carotene oxydases. The carotene-bleaching activity of the pH 6,5 lipoxygenases was inhibited by NDGA. During the test on carotene oxydases activity the carotene was not oxidized by the linoleic acid hydroperoxides.
Zusammenfassung
Extrakte aus Erbsen, Sojabohnen, Weizen und Leinsamen wurden durch Gel- oder Ionenaustauschchromatographie getrennt. Die Bestimmung der Carotin-Abbau-Aktivität der Eluate ergab: Die Lipoxygenase-Isoenzyme mit einem pH-Optimum bei 6,5 sind sehr wirksame Carotinoxydasen, wenn Linolsäure im Incubationsansatz vorhanden ist. Solche Carotinoxydasen kommen in Erbsen, Sojabohnen und im Weizen vor. Im Unterschied dazu bauen die alkalische Lipoxygenase aus Sojabohnen, die Hydroperoxidisomerase aus Leinsamen und die Peroxidasen aus Weizen und Sojabohnen nur sehr langsam das Carotin ab. Gehemmt wird die Carotin-Bleichung der pH 6,5-Lipoxygenasen durch NDGA. Linolsäurehydroperoxide oxydieren nicht das β-Carotin während der Bestimmung der Carotinoxydase-Aktivität.
