Summary
An HPLC method is described for the separation and determination of 16 Amadori compounds in dried vegetables and other vegetable products. Fructose, glucose and fructose-pyrrolidonic acid, the cyclization product of fructoseglutamic acid and fructoseglutamin, can also be detected by this method. After extraction of the samples with water, separation is performed by isocratic HPLC on DEAE silica gel using an acetonitrile phosphate buffer mobile phase. A post-column reaction with an alkaline solution of triphenyltetrazoliumchloride at 85 °C allows a very selective and sensitive photometric determination at 480 nm. Detection as low as 10–50 ng for the various Amadori compounds can be achieved. The analysis of a number of industrially dried vegetables led to concentrations of 0.01 %–3.8% (in dried matter) for the individual compounds. The highest amounts of Amadori compounds were found in dried tomatoes, dried paprika and dried asparagus since the raw products contain high amounts of glucose and free amino acids, which are the precursors of the Amadori compounds.
Zusammenfassung
Es wird eine Methode beschrieben, mit der gleichzeitig 16 Amadori-Verbindungen in Trockengemüse und Gemüseprodukten bestimmt werden können. Zusätzlich werden Fructose, Glucose und Fructosepyrrolidoncarbonsdure, ein Cyclisierungsprodukt von Fructoseglutamin und Fructoseglutaminsäure, erfaßt. Nach Extraktion mit Wasser aus dem Untersuchungsmaterial erfolgt die isokratische HPLC-Trennung der Amadori-Verbindungen mittels Acetonitril/Phosphatpuffer an einer DEAE-Kieselgelsäule. Der empfindliche und spezifische
Nachweis der getrennten Verbindungen erfolgt durch post-column Umsetzung mit alkalischer Triphenyltetrazoliumchloridlösung bei 85 °C und anschließender Detektion des gebildeten Triphenylformazans bei 480 nm. Die Nachweisgrenzen liegen bei 10 ng bis 50 ng für die verschiedenen Verbindungen. Mit dieser Methode wurden zahlreiche industriell hergestellte Trockengemüse untersucht und für die einzelnen Amadori-Verbindungen Gehalte zwischen 0,01% und 3,8% i. T. ermittelt. Besonders hohe Gehalte fanden sich bei Tomaten-, Gemüsepaprika- und Spargeltrokkenprodukten, da hier in den Ausgangsprodukten relativ hohe Gehalte an Glucose und freien Aminosäuren als Vorstufen der Amadori-Verbindungen vorhanden sind.
