Summary
Oxygen dissociation curves of diluted haemoglobin solutions are plotted with the aid of a test procedure, outlined by Thews (1960), for the examination of O2 diffusion in biological media. The applicability of this process for the direct and exact determination of oxygen dissociation curves is verified both theoretically and in practice. Tests on greatly diluted human haemoglobin solutions, of concentration 16.4×10−3 g/100 ml and 16.4×10−4 g/100 ml, give the following results:
At temperatures of 20, 30, and 37°C, with a pH value of 8.0, the oxygen dissociation curves of the haemoglobin solutions tested, display typical slopes. The oxygen affinity of the test-solutions increases with diminishing concentration.
Zusammenfassung
Mit Hilfe einer von Thews (1960) für Untersuchungen der O2-Diffusion in biologischen Medien angegebenen Versuchsanordnung werden Sauerstoffverbindungskurven von verdünnten Hämoglobinlösungen aufgenommen. Die Anwendbarkeit des Verfahrens für die direkte und genaue Bestimmung von Sauerstoffbindungskurven wird theoretisch und praktisch überprüft. Untersuchungen an hochverdünnten Lösungen menschlichen Hämoglobins einer Konzentration von 16,4·10−3 g/100 ml und 16,4·10−4 g/100 ml führen zu folgenden Ergebnissen:
Bei den Temperaturen 20, 30 und 37°C und dem pH-Wert 8,0 haben die Sauerstoffbindungskurven der untersuchten Hämoglobinlösungen typisch S-förmige Verlaufsformen. Die Sauerstoffaffinität der Untersuchungslösungen steigt mit abnehmender Konzentration.
