Experimental Methods for Evaluating Pharmacodynamic Effects of Drugs for the Pulmonary System

Abstract

Asthma and chronic obstructive pulmonary disease (COPD) are distinct respiratory diseases with unmet medical needs affecting hundreds of millions of patients worldwide. While asthma and COPD do share a number of common features as they are chronic inflammatory conditions of the lung, they are clearly different diseases with unique characteristics and causes. However, treatment of both these diseases remains primarily the same. There is clearly a need for different drug classes to be developed for the treatment of these conditions. The use of animal models in investigating potential new drugs for asthma has tried to replicate both the early and late asthmatic response to allergen challenge, bronchial hyperresponsiveness, and airway remodelling. Historically, the guinea pig was the species of choice with its many advantages. However, more recently mice have been more widely used in preclinical research although they lack many features of the human condition. The primary characteristic of COPD that is replicated in animal models is non-allergic inflammation. The main challenge with this is that COPD often develops over a significant period of time which is difficult to accurately replicate in an animal model. Once again, different species have been used to best investigate the various aspects of this inflammation. In addition, cough is also an important feature of chronic lung conditions for which no treatment has been approved. Establishing improved animal models that exhibit a hypertussive response would be essential in the development of antitussive drugs. Overall, there is a clear need for the development of improved animal models to aid in the discovery and development of new treatments in both asthma and COPD.
