Abstract
The automatic stepwise degradation of polypeptides with phenylisothiocyanate was introduced almost 20 yr ago (1). This invention has greatly enhanced the effectiveness of the sequential degradations, increased the repetitive yields, lowered the background contaminations of the released amino acid derivatives, and allowed considerably extended degradation of polypeptides in comparison to any of the manual techniques available at that time. This so-called liquid-phase technique, in which the protein is degraded as thin film in a spinning cup, has been widely used and successfully applied to the sequence determination of proteins and polypeptide fragments.
