In isole pancreatiche ottenute da topi obesi-iperglicemici è stata studiata la captazione di3H-tolbutamide. Il farmaco viene captato in eccesso rispetto allo spazio extracellulare delle isole. La captazione è saturabile, con massimo di circa 3 mmoli/kg di peso secco e valore semimassimo ad una concentrazione 2 mM nel mezzo. Albumina e glibenclamide deprimono la captazione della tolbutamide da parte delle isole. Questi dati confermano l’opinione secondo la quale la tolbutamide stimolerebbe la liberazione insulinica mediante formazione di Iegami specifici con la membrana plasmatica delle cellule B.
Résumé
On a étudié l’absorption de3H-tolbutamide par les ilôts pancréatiques de souris obèses hyperglycémiques. Le médicament est absorbé en excés relativement à l’espace extracellulaire des ilôts. L’absorption est saturable, avec un maximum d’environ 3 mmoles/kg de poids sec, et avec une valeur semi-maximum à la concentration 2 mM dans le milieu. Albumine et glibenclamide dépriment l’absorption de tolbutamide de la part des ilôts. Ces données confirment l’opinion selon laquelle le tolbutamide stimulerait la libération insulinique au moyen de la formation de liaisons spécifiques avec la membrane plasmatique des cellules B.

Se ha estudiado la captación de3H-tolbutamida en islas pancreáticas procedentes de ratones obeso-hiperglucémicos. Respecto al espacio extracelular de las islas, el fármaco resulta captado en exceso. La captación se puede saturar con un máximo de unos 3 mmoli/kg de peso seco y valor semimáximo a la concentración 2 mM en el medio. La albumina y la glibenclamida deprimen la captación de la tolbutamida por parte de las islas. Estos datos corroboran la opinión según la cual la tolbutamida estímula la liberación de insulina por medio de la formación de vínculos específicos con la membrana plasmática de las células B.
Zusammenfassung
Die Aufnahme von3H-markiertem Tolbutamid seitens der Pankreasinseln von adipös-hyperglykämischen Mäusen wurde untersucht. Die Substanz wird in einem Raum aufgenommen, der grösser ist als der extrazelluläre Raum der Inseln. Die aufnhame hat einen Sättigungspunkt und erreicht ihr Maximum bei 3 mMol/kg Trockengewicht und ihren Halbwert bei einer Konzentration von 2 mM im Medium. Albumin und Glibenclamid deschränken die Tolbutamidaufnahme. Diese Befunde bestätigen die Ansicht, dass Tolbutamid die Insulinfreisetzung mittels Bildung spezifischer Bindungen an der Plasmamembran der B-Zelle fördert.
Summary
The uptake of3H-labelled tolbutamide by microdissected pancreatic islets from obese-hyperglycemic mice has been studied. Tolbutamide is taken up in excess of the extracellular space of the islets. The uptake is saturable with a maximum uptake of about 3 mmoles/kg dry weight and a half-maximum at a 2 mM concentration in the medium. Albumin and glibenclamide depress the islet uptake of tolbutamide. The data support the view that tolbutamide stimulates insulin release by specific binding to the plasma membrane of the B-cells.
