Summary
A method for quantitative evaluation of phototactic experiments is described which is especially suitable for Cyanophyceae of theOscillatoria-Phormidium type. The zero threshold for topotaxis ofPhormidium autummale is at 5 lux, the reaction optimum at 200 lux, and the maximum, i. e. region of indifference, is reached at 10000 lux. Negative topotaxis was not observed, since the movement ofPh. autumnale ceases above 10000 lux as a result of negative photokinesis.
With the aid of an interference filter monochromator system the action spectrum for topotaxis was determined for the range between 317 and 800 mμ at a constant energy of 1000 erg/cm2·sec. These results were correlated with studies on the pigment content.
Three main maxima were found at 395–405, 490–500 and 560 to 570 mμ. The second maximum corresponds to the principle absorption region of the carotenoids and the last to that of the phycoerythrin. In agreement with the low phycocyanin content a small subsidiary maximum at 615 mμ was found. Thus visible irradiation is photo-topotactically active in proportion to the carotenoid and phycobilin absorption, whereas apparently no connection exists with chlorophyll a. Possible interpretations of the maximum at 395–405 mμ are discussed.
