Riassunto
Oggetto del presente lavoro è la secrezione insulinica da parte di colture monostratificate di tessuto pancreatico di ratti adulti e di maiali neonati. La quantità di ormone liberata diminuisce con l'aumentare dell'età delle cellule in accrescimentoin vitro. Il contenuto in insulina del mezzo nutritivo è una funzione dell'ormone secreto e di quello metabolizzato dalle cellule. La quantità di insulina liberata durante il periodo della crescitain vitro dipende dalla frequenza con cui il mezzo nutritivo viene rinnovato. L'aggiunta di insulina eterologa al mezzo di coltura influenza — cioè inibisce — la liberazione insulinica, a condizione che le cellule consumino l'ormone ad un ritmo costante. Secondo la nostra opinione, le concentrazioni inibitrici dipendono dallo stato metabolico delle cellulein vitro.
Résumé
Cette étude a pour objet d'étudier la secrétion insulinique par des cultures monostratifiées de tissu pancréatique chez des rats adultes et des porcs nouveau-nés. La quantité d'hormone dégagée diminue à mesure qu'augmente l'âge des cellules croissantin vitro. La teneur en insuline dans le milieu nutritif est fonction de l'hormone secrété et de l'hormone métabolisé par les cellules. La quantité d'insuline libérée pendant la croissancein vitro tient à la fréquence avec laquelle le milieu nutritif est renouvelé. L'addition d'insuline hétérologue au milieu de culture influence — inhibe — le dégagement de l'insuline, à la condition que les cellules utilisent l'hormone à un rythme constant. A notre avis, les concentrations inhibitoires dépendent de l'état métabolique des cellulesin vitro.
Resumen
El objeto del presente trabajo es el estudio de la secreción insulínica por parte de cultivos monoestratificados de tejido pancreático de ratas adultas y de suinos recién nacidos. La cantidad de hormón liberado disminuye con el aumento de la edad de las células en desarrolloin vitro. El contenido de insulina del medio nutritivo es una función del hormón segregado y del metabolizado por las células. La cantidad de insulina liberada durante el período de crecimientoin vitro depende de la frecuencia con que se renueva el medio nutritivo. La agregación de insulina heteróloga respecto al medio de cultivo influencia — es decir, inhibe — la liberación insulínica, a condición de que las células vayan consumiendo el hormón a un ritmo constante. Según nuestra opinión las concentraciones inhibitorias dependen del estado metabólico de las célulasin vitro.
Zusammenfassung
Objekt der gegenwärtigen Untersuchung ist die Insulinsekretion seitensMonolayer-Kulturen aus Pankreasgewebe von erwachsenen Ratten und neugeborenen Schweinen. Die Menge an freigesetztem Hormon nimmt mit zunehmendem Alter derin vitro wachsenden Zellen ab. Der Insulingehalt des Nährmittels steht in Bezug zum sezernierten und zu dem von den Zellen metabolisierten Hormon. Die freigesetzte Insulinmenge während der Wachstumsphasein vitro hängt von der Frequenz ab, mit der das Nährmittel erneuert wird. Der Zusatz von heterologem Insulin zum Kulturmittel beeinflusst — d.h. hemmt — die Insulinfreisetzung unter der Bedingung dass die Zellen das Hormon mit konstantem Rhythmus verbrauchen. Nach unserer Meinung hängen die hemmenden Konzentrationen vom metabolischen Zustand der Zellenin vitro ab.
Summary
This article deals with the insulin secretion of monolayer cultures of pancreatic tissue of adult rats and newborn pigs. The amount of insulin released diminishes with increasing age of the cells growingin vitro. The insulin content of the nutrient medium is a function of the insulin released and the insulin metabolised by the cells. The amount of insulin yielded during the growth periodin vitro depends on the frequency with which the nutrient medium is changed. Adding heterologous insulin to the culture medium influences, i.e. inhibits, insulin release, provided the cells consume insulin at a constant rate. The inhibitory concentration (we assume) depends on the metabolic condition of the cellsin vitro.
