Summary
Sulfaphenazol potentiates the hypoglycaemic effect of tolbutamide by prolonging its half-life in plasma. The attempt has been made in man to localize the step in tolbutamide metabolism which is inhibited by sulfaphenazol. It was found to prolong the half-life of undegraded tolbutamide without interfering with the elimination of its metabolites hydroxy-tolbutamide and carboxy-tolbutamide. It seems that sulfaphenazol blocks the first stage of tolbutamide metabolism, its hydroxylation, whilst the second step — carboxylation — remains uninhibited and is accomplished in a few minutes.
