Aus Erd-und Schlammproben wurden zehn fakultativ chemolithotroph wachsende Hydrogenomonas-Stämme angereichert und isoliert.

Bei chemolithotroph wachsenden Zellen nimmt das Zellgewicht während der exponentiellen Wachstumsphase fortlaufend ab. Bei Rührung der Nährlösung mit 650 U/min erfolgt die Zellvermehrung unter einer Atmosphäre von 65% H2+5% CO2+30% O2 von 4·107 bis 2·109 Zellen/ml exponentiell. Während die Zellzahl in der stationären Phase annähernd konstant bleibt, nimmt die Trübung der Suspension infolge Speicherstoffsynthese (Poly-β-Hydroxybuttersäure) weiterhin zu.

Die Geschwindigkeit der H2-Aufnahme durch gewaschene, chemolithotroph herangezogene Zellen ist in Abwesenheit von Kohlendioxyd mit Methylenblau als H-Acceptor etwa dreimal so hoch wie mit Sauerstoff. Unter einem Gemisch von H2+O2 erhöht CO2 die Rate der H2-Oxydation ebenfalls auf das Dreifache der CO2-freien Kontrolle. Unter denselben Bedingungen steigern zugesetzte organische Säuren (Brenztraubensäure, β-Hydroxybuttersäure) die Rate der H2-Aufnahme maximal um nur 30%.

Gewaschene, chemolithotroph gewachsene Zellen assimilieren organische Substrate oxydativ und bauen Poly-β-Hydroxybuttersäure auf; die Syntheserate ist mit Hydroxybuttersäure, Milchsäure und Essigsäure am höchsten. Während die Syntheserate bei den meisten organischen Substraten durch gleichzeitige H2-Oxydation gesteigert wird, wird die Synthese aus Essigsäure und Hydroxybuttersäure durch H2 gehemmt. In Gegenwart organischer Säuren verläuft die autotrophe CO2-Fixierung langsamer als ohne organische Substrate.
