Abstract
The in vivo Matrigel angiogenesis assay, or Matrigel plug assay, was developed to examine neovascularization in rodents in response to specific angiogenic factors, independently of the presence of tumor cells (Kibbey et al. 1992, Passaniti et al. 1992, Grant et al. 1993, Capogrossi and Passaniti 1999). Since its inception, a variety of agents have been used to deliver an angiogenic stimulus, including recombinant cytokines, adenoviral vectors, low dose tumor cells, and even telomerase-transfected endothelial cells (Muhlhauser et al. 1995, Klement et al. 2000, Akhtar et al. 2001, Yang et al. 2001). The applications of the assay include (1) the study of the basic mechanisms underlying angiogenesis at the molecular or cellular level and (2) the in vivo evaluation of the activities of angiogenic and angiostatic compounds or cytokines (Capogrossi and Passaniti 1999).
