Abstract
In Escherichia coli, different translational initiation sites can vary 1000-fold in their intrinsic efficiencies.1,2 These differences are attributable to specific sequence features that are involved in ribosome-messenger (mRNA) interactions as well as to mRNA secondary structures that can inhibit ribosome binding. In addition to the variation in intrinsic strengths of different initiation sites, some sites are subject to regulation so that their efficiencies are modulated according to the state of the cell. For the purposes of this chapter, translational regulation means the action of a gene product that affects the rate of translational initiation for some protein. Other posttranscriptional effects on rates of translation, such as differential mRNA degradation and translational coupling, are not included.
