Abstract
Digitonin treatment of chloroplasts is widely used to obtain PS I and PS II particles in concentrations higher than 1 per cent. The digitonin influence (below 1 per cent) on the proton uptake, FeCN photoreduction and on the behaviour of spin probes in chloroplast membranes is described in this study. Glutaraldehyde (GA) is a cross-linking reagent for soluble and membrane bound proteins and makes inter- and intramolecular bonds. GA treated chloroplasts preserve the partial ability for photoinduced transmembrane proton transport, O2 evolution, photoreduction of FeCN, lose the photophosphorylation activity and are osmotically inert. The influence of a GA on the molecular organization is not quite clear.
