Genetically Expressed Monodisperse α Helical Polypeptides

Abstract

Four different chain lengths of monodisperse, α-helical polypeptides derived from the protein comprising the tail fibers of the bacteriophage Ur-λ were prepared by recombinant DNA techniques. The DNA which codes for the tail fiber segment has been replicated and inserted into Escherichia Coli bacteria. The desired proteins expressed by the bacteria were harvested and purified with a nickel affinity chromatography. The target proteins were characterized by gel electrophoresis and matrix-assisted laser desorption/ionization (MALDI) mass spectrometry. The secondary α-helical structure of the polypeptides was characterized by circular dichroism (CD) spectrophotometry.
