Abstract
The hepatic very low density lipoprotein (VLDL) synthesis seems to depend not only on the concentration of substrates affluent to the liver, but on the nutritional and hormonal milieu as well. It has long been known that changes in the circulating levels of insulin and glucagon can shift glucose metabolism in the liver from storage to release. It has been recognized only recently that this same hormone couple also plays a pivotal role in hepatic fat metabolism (Eaton 1977; Mc Garry 1979). Glucagon and insulin exert opposite effects upon lipid metabolism both in peripheral tissues and in liver (Fig. 1). Insulin and glucagon have to be secreted, and transported to target tissues, in which they must interact with specific receptors to initiate a series of postreceptor events. Defects in any of these processes can impair lipoprotein metabolism.
