Summary
Occlusion of the middle cerebral artery (MCA) in cats results in a drop in blood flow in the territory normally supplied by the MCA, demonstrable by the measurement of rCBF with 85Krypton or by angiography. As a consequence of tissue hypoxia a rise in lactate, in lactate/pyruvate ratio and a breakdown of CrP, ATP and the energy charge potential is found in the ischemic area. At the borderline zone a slight hyperemia, followed by a secondary depression in blood flow, was recorded, accompanied by comparable alterations in energy-rich phosphates and in L/P ratio. A significant increase in water content of the grey and white matter occured already after 30 or 120 min, respectively. The accumulation of edema fluid takes place in the absence of protein extravasation.
A group of cats were pre-treated with dexamethasone (0.3 mg/kg/day; intraperitoneally), starting 72 h before the occlusion. In dexamethasone-treated animals the accumulation of fluid in the ischemic as well as in the bordering zone was diminished 2 h after the occlusion as compared to untreated animals. rCBF changes were now less pronounced and the energetic situation was better preserved.
