Abstract
Before 1956 two “facts” were known about human cytogenetics. The human chromosome number was believed to be 48, and the XX-XY mechanism of sex determination was assumed to work in the same way as it does in Drosophila. Studies of the fruit fly had shown that the ratio of the number of X chromosomes to the number of sets of autosomes determines the sex of the organism. Both of these fundamental notions about human chromosomes were eventually proved wrong.
