Abstract
Numerous teratological investigations have provided evidence that chemical or physical insult to the developing embryo often produces within a few hours or days obvious signs of cell necrosis in tissues destined to be malformed. This observation has been made so often that one hardly need hesitate in ascribing the malformation to the earlier cytotoxicity. However, further observations cloud the issue somewhat: (1) Agents which at high doses produce cell death and teratogenesis, at low doses continue to kill cells (albeit at a lower rate) but may not produce malformations, and (2) many cytotoxic, teratogenic agents produce cell death in tissues which appear normal at birth. This means that cell death above and beyond physiological levels does not inevitably lead to malformation. Thus, it becomes necessary to know the secondary consequences of cell death in a particular organ system so that the temporal and quantitative aspects of necrosis needed to produce malformation become explainable.
