Gli AA. hanno studiato gli effetti del 2-desossiglucosio (2-DG) sulla morfologia delle cellule insulari e sui livelli di insulina nel sangue portale. Le cellule B non hanno mostrato alcuna alterazione della loro ultrastruttura, se si eccettua un aumento dei corpi densi. Degranulazione è stata osservata, in alcune di queste cellule, 6 h dopo la somministrazione del 2-DG. Dopo 45 min dalla iniezione del 2-DG, i livelli insulinemici apparivano bassi, nonostante la notevole iperglicemia; dopo 6 h, essi erano lievemente superiori alla norma. Résumé
On a étudié l’effet du 2-desoxyglucose (2-DG) sur la morphologie des cellules insulaires et sur le niveau de l’insuline dans le sang portel. Les cellules B n’ont présenté aucune altération de leur ultra-structure, à l’exception d’une augmentation des corps denses. Six heures après l’administration de 2-DG, une dégranulation a été observée dans quelques cellule. Quarante-cinq minutes après l’administration, la concentration de l’insuline était faible, malgré la forte hyperglycémie; après six heures elle était à peine inférieure à la normalité. Les cellules A sont restées toujours bien granulées. Les cellules B ont présenté des signes d’activité fonctionnelle, comme, par example, la formation de microvillosités, l’augmentation de l’ergastoplasme lamellaire et du nombre des ribosomes libres. On discute la signification de ces résultats.
Resumen
Los autores han estudiado los efectos de la 2-deoxiglucosa (2-DG) en la morfología de las células insulares y en los niveles de insulina de la sangre portal. Las células B, si se exceptúa un aumento de los cuerpos densos, no han presentado ninguna alteración en su ultraestructura. En algunas de esas células se observó degranulación a las 6 horas del suministro del 2-DG. A los 45 minutos de la inyección de 2-DG, los niveles insulinémicos aparecían bajos a pesar de la notable hiperglucemia y después de las 6 horas ya se habían elevado levemente por encima de la normalidad. Las células A seguían bien granuladas durante toda la duración del experimento. Las células D acusaron indicios de actividad funcional, tales como la formación de microvellosidades, el aumento de ergastoplasma lamellar y de los ribosomas libres. Zusammenfassung
Es wurde die Wirkung von 2-Deoxyglukose (2-DG) auf die Morphologie der Inselzellen und auf den Insulinspiegel im Pfortaderblut untersucht. Die B-Zellen zeigten keinerlei Veränderungen ihrer Feinstruktur, wenn man von einer Zunahme der dichten Körper absieht. Einige B-Zellen zeigten 6 Stunden nach Verabreichung von 2-DG Degranulationserscheinungen. Trotz ausgesprochener Hyperglykämie war der Insulinspiegel 45 min nach 2-DG-Verabreichung niedrig, nach 6 Stunden dagegen war er etwas der Norm gegenüber erhöht. Die A-Zellen blieben nach 2-DG durchwegs normal granuliert. Die D-Zellen wiesen Zeichen von Aktivität auf, nämlich Bildung von Mikrovilli, vermehrtes lamelläres Ergastoplasma und freie Ribosomen. Die Bedeutung dieser Befunde wird besprochen.
Summary
The effect of 2-deoxyglucose (2-DG) on islet cell morphology and portal vein insulin levels was studied. The B-cells did not show any alteration in their fine structure, except for an increase in dense bodies. Six hrs after 2-DG administration degranulation was observed in some of these cells. Insulin levels were low 45 min after 2-DG administration, in spite of the marked hyperglycemia, and slightly higher than normal after 6 hrs. The A-cells remained well granulated at all times after 2-DG injection. The D-cells showed signs of functional activity, such as microvilli formation, increased lamellar ergastoplasm and free ribosomes. The significance of these findings is discussed.
