Zusammenfassung
Durch Colchicinbehandlung haploider Kartoffelsamen wurden unter 86 vollständig analysierten Pflanzen 50% total verdoppelten und 23% Chimären (24-24-48, 24-48-48, 48-24-24, 48-24-48, 96-48-48, und 96-48-96 in L I, L II und L III) gewonnen. Die Häufigkeiten der Ploidieerhöhung betrugen: L I: 69,8%>L III: 65,1%>L II: 60,5%. Sie folgen nicht der Zufallsverteilung. L I und L II differierten häufiger als L II und L III.
Die Versuche zeigen, dass alle drei Initialzellschichten schon im Keim reifer Kartoffelsamen differenziert sind.
Beim Übergang von haploider zu diploider Stufe betrug der Verdopplungsfaktor für Schliesszellenplastidenzahlen 1,86 (22,73∶12,20). Er sank beim Übergang von 48 auf 96 Chromosomen auf 1,74 (39,46∶22,73) herab.
Der Pollen relativ homozygoter, colchicinverdoppelter Haploider ist grösser und hat bedeutend höhere Keimporenzahlen (28,85±0,26 μ; 3,88±0,05 Poren) als der Pollen stark heterozygoter, normaler Kartoffelzuchtstämme (28,24±0,48 μ; 3,41±0,05 Poren).
Durch Chromosomenverdopplung ging die Pollenfertilität generell stark zurück (91% der Haploiden und 23% der Diploiden überschritten 40% Färbbarkeit).
Summary
In a colchicine-doubling experiment with 377 haploid potato seeds from two reciprocal crosses, 111 seedlings were raised to maturity and 86 of them flowered (Table 1, cf. Fig. 2). Of these latter, 23 plants were unchanged haploids, 43 (50%) were totally doubled and 20 (23.3%) were ploidy chimeras. Among the 25 non-flowering plants at least 5 ploidy chimeras were found. The total number of ploidy chimeras amounted to at least 22.5% (Table 2).
Among the chimeras, the following combinations of the three germ layers, L I, L II and L III, were identified: 24-24-48, 24-48-48, 48-24-24, 48-24-48, 96-48-48, 48-48-96 and 96-48-96 (Table 2).
Among the 258 germ layers from 86 completely analysed plants, 65.1% were doubled or quadrupled. L I showed the highest frequency of ploidy increase (69.8%), followed by L III with 65.1%. The smallest frequency: 60.5%, was found in L II.
The germ layers were not chromosome doubled according to a random distribution scheme: a simultaneous doubling of all three layers frequently occurred (Table 3). A difference in the ploidy levels of L I and L II occurred a significantly higher number of times than in the levels of L II and L III.
From the results, it is concluded that all three germ layers are already present in the epicotyl apex of the embryo from mature potato seeds. Various concepts of epicotyl structure are discussed.
Ploidy chimeras with 96 chromosomes in L II only, or in L III only, are unstable.
In some cases, the isolation of one chimera partner was carried out, using the eye-excision method. The doubling factor for the chloroplast number of the guard cells was 1.86 for the transition from the 24 to the 48-chromosome level (chloroplast numbers 12.20±0.175 and 22.73±0.272 respectively). For the transition from the 48 to the 96 chromosome level (chloroplast number 39.46±0.67), it was somewhat lower, viz. 1.74 (Fig. 3). The pollen diameter of colchicine-doubled haploids, 28.85±0.26 μ, is significantly larger than that of normal potato breeding lines: 28.24±0.48 μ. The germ pore number of the former: 3.88±0.05, is also very significantly higher than that of the latter: 3,41±0.05. These differences are ascribed to the strong degree of homozygosity of colchicine-doubled haploids (Table 4). The excellent pollen fertility of the original haploid material (91% of the lines with more than 40% stainable pollen) is, after doubling, generally considerably reduced (only 23% of the lines with more than 40% stainable pollen, cf. Fig. 1).
Résumé
Lors d'un essai de doublement de chromosomes par traitement à la colchicine de 377 semences de Pommes de terre haploïdes provenant de deux croisements réciproques, 111 plantules ont atteint la maturité dont 86 ont fleuri (Tableau 1, cf. Fig. 2). Parmi ces dernières plantes, 23 étaient restées haploïdes, 43 (50%) étaient totalement doublécs et 20 (23,3%) étaient des chimères ploïdiques. Il y avait au moins 5 chimères ploïdiques parmi les 25 plantes ne fleurissant pas. Le nombre total de chimères ploïdiques atteignait au moins 22,5% (Tableau 2).
Parmi les chimères, on a identifié les combinaisons suivantes dans les trois couches de cellules apicales L I, L II et L III: 24-24-48, 24-48-48, 48-24-24, 48-24-48, 96-48-48, 48-48-96 et 96-48-96 (Tableau 2).
Parmi les 258 couches de cellules apicales des 86 plantes complètement analysées, 65,1% étaient doublées ou quadruplées. L I montrait la plus haute fréquence d'augmentation ploïdique (69,8%), suivi par L III avec 65,1%. La plus basse fréquence (60,5%) était trouvée en L II. Les doublements chromosomiques dans les couches de cellules apicales ne suivaient pas un schéma de répartition au hasard: fréquemment un doublement des trois couches apparaissait simultanément (Tableau 3). Significativement plus élevée était le nombre de fois où apparaissait une différence des niveaux ploïdiques de L I et L II par rapport à L II et L III.
Les résultats permettent de conclure que les trois couches de cellules apicales sont toujours présents dans le sommet épicotyle de l'embryon de semences mûres de Pomme de terre. L'auteur discute de différentes conceptions de la structure de l'épicotyle.
Les chimères ploïdiques avec 96 chromosomes en L II seulement, ou en L III seulement, sont instables.
Dans quelques cas l'isolation de chimères associées a été réalisé par la méthode d'excision d'yeux.
Le nombre de chloroplastes des cellules stomatiques augmente suivant le coefficient 1,86: le nombre de chloroplastes est de 12,20±0,175 au niveau haploïde et de 22,73±0,272 au niveau 48 chromosomes. Du dernier niveau mentionné au stade 96 chromosomes (nombre de chloroplastes: 39,46±0,67) le coefficient d'augmentation de doublement 1,74 est légèrement réduit (Fig. 3).
Le diamètre du pollen des haploïdes doublés à la colchicine, 28,85±0,26 μ, est significativement plus grand que celui des souches normales d'amélioration: 28,24±0,48 μ. Le nombre de pores du germe du premier groupe: 3,88±0,05, est très significativement plus élevé que celui des clones normaux de pomme de terre: 3,41±0,05. Ces différences sont attribuées au fort degré d'homozygotie des haploïdes doublés à la colchicine (Tableau 4).
Généralement, l'excellente fertilité pollinique du matériel haploïde original (91% des lignées avec plus de 40% du pollen se colorant) est, après doublement, considérablement réduit (seulement 23% des lignées ont plus que 40% du pollen se colorant, cf. Fig. 1).
