Zusammenfassung
Die von Natti (1955) beschriebene Verringerung in der Quellbarkeit der Stärke virusinfizierter Kartoffelknollen, die nach Zugabe von verdünnter Säure in Erscheinung tritt, konnte reproduziert werden. Die Stärkequellung wurde jedoch in so starkem Maße von kleinen Abweichungen bei der Bemessung von Gewebebrei und Säuremenge beeinflußt, daß sie in dieser Versuchsanordnung kaum zu virusdiagnostischen Zwecken geeignet war. Im Verlauf von Untersuchungen zur Verbesserung der Versuchsanordnung waren die angeführten Schwankungen beträchtlich verringert, wenn die Stärkequellung nicht in Säure, sondern in erwärmten Stärkesuspensionen erfolgte.
Das als Stärkequellungstest bezeichnete Verfahren wird beschrieben. Auf Grund von methodischen Untersuchungen wird die zweckmäßigste Versuchsanordnung festgelegt. Erste Untersuchungen grösseren Umfanges, die zur Kennzeichnung der Arbeitssicherheit durchgeführt wurden, ergaben, dass befriedigende Korrelationen zwischen den an einer größeren Knollenzahl erhaltenen durchschnittlichen Testergebnissen und dem durchschnittlichen Virusbesatz einer Herkunft bestehen. Werden jedoch die an einzelnen Knollen gewonnenen Testergebnisse unmittelbar mit dem im Stecklingstest bzw. im Feldanbau der geprüften Knollen ermitteltem Gesundheitszustand verglichen, so treten mitunter größere Differenzen auf, die der Anwendung des Testes gewisse Grenzen setzen dürften.
Summary
The decreased ability of starch from virus infected tubers to swell when treated with dilute acid, described by Natti (1955), was confirmed. The swelling was, however, greatly influenced by small differences in the measurement of the quantities of tissue pulp and acid and was of little use in the diagnosis of virus infection. In the course of experiments to improve the technique it was found that such variations were markedly reduced when the swelling was induced in a warmed starch suspension rather than in acid. Using this procedure for a starch swelling test, tissue pulp was taken from the tubers under investigation and placed in water for 15 minutes at a temperature specific to the variety. After staining with iodine in potassium iodide, the proportion of swollen starch grains was then determined microscopically, using polarised light. The result was expressed on a scale (Wz) of from 1 to 5, 1 denoting all grains in a test swollen and 5 virtually none. In a series of experiments a number of factors were examined in relation to their influence on the results of the test. This showed, amongst other things, that the ability of the starch to swell could best be demonstrated on starch grains from the rose-end of the tuber (Table 1). Preliminary large scale experiments, carried out to determine the reliability of the test, showed that in many varieties it is satisfactory provided that the mean obtained from a large number of tubers is set against the mean virus infection of material from the same stock (Table 2). Results from individual tubers could not be related directly to the health status of the same tubers (Table 3). When results from all the varieties are taken into consideration, the difference between healthy and virus-infected tubers in the ability of their starch to swell can be shown to be statistically highly significant. With individual varieties, however, the level of agreement is variable and there can be large differences which must set distinct limits to the use of the tests.

La diminution, décrite par Natti, de l'aptitude au gonflement de l'amidon chez les tubercules de Pomme de terre infectés de virus, qui se manifeste après apport d'un supplément d'acide dilué, peut être reproduite. Le gonflement de l'amidon était toutefois influencé par le grand volume des petites différences dans la mensuration de la bouillie de tissu et de la quantité d'acide, de sorte qu'il n'était presque pas utilisé dans l'examen en série pour le diagnostic des viroses. Au cours des recherches pour améliorer les examens en série, les variations rapportées ont été considerablement diminuées quand le gonflement de l'amidon n'avait pas leiu en milieu acide, mais en suspensions chauffées. Pour l'exécution des procédés utilisés comme tests de gonflement de l'amidon, on utilisera de la bouillie de tissu des tubercules examinés qui aura été placée dans l'eau durant 15 min. à une température spécifique pour la variété. Ensuite on déterminera, par la coloration au iodure de potassium en lumière polarisée sous microscope, quelle partie des grains d'amidon contenus dans la bouillie de tissu est gonflée. Cette partie sera exprimée en cotes de 1 à 5. A la cote 1 tous les grains d'amidon d'un test sont gonflés, tandis qu'à la cote 5 il n'y en a aucun. Dans des déterminations méthodiques, on a testé un certain nombre de facteurs de la série de recherches quant à leur influence sur les résultats. C'est ainsi qu'il apparut, notamment, que les différences dans l'aptitude au gonflement de l'amidon se manifestait le mieux chez les grains d'amidon de la couronne (Tableau 1). Les premières déterminations des plus grands diamètres exécutées pour préciser le degré de sécurité du travail, révélaient que celui-ci était suffisant pour un grand nombre de variétés, pour autant que le résultat du test de la teneur moyenne en virus s'applique à un grand nombre de tubercules moyens mais d'une origine déterminée (Tableau 2). Les résultats de test obtenus à partir de tubercules isolés étaient comparés directement avec l'état de santé réel des tubercules correspondants (Tableau 3), les différences trouvées entre tubercules sains et tubercules infectés de virus dans le gonflement de l'amidon étaient parfaitement confirmées statistiquement quand ces résultats étaient obtenus sur toutes les variétés. Pour quelques variétés cependant le degré de concordance varie grandement et de grosses différences apparaissent, qui peuvent poser certaines limites à l'emploi du test.
