Zusammenfassung
Bei einem Neugeborenen wurde am 7. Lebenstage eine Galaktosurie und Hypergalaktosämie gefunden, die durch Mangel an Galaktokinase bedingt ist. Radiochromatographisch konnte an Erythrocyten-Hydrolysat gezeigt werden, daß die Bildung von phosphorylierten Galaktosederivaten aus Galaktose-l-14C auf etwa 4% und die Bildung von14CO2 ebenfalls stark herabgesetzt war. Die Galaktose-l-P-Uridyltransferase erwies sich im optischen Test als normal aktiv.
Vor Beginn einer milchfreien Diät im Alter von 5 Wochen entstanden Katarakte, die sich später zurückbildeten. Die Heredität dieser Erkrankung ist anzunehmen, da die Eltern blutsverwandt und zwei Sippenangehörige als Kinder erblindet sind. Auffällig ist die Häufung dieses Enzymdefekts bei Zigeunern.
Summary
A newborn with hypergalactosaemia and galactosuria due to galactokinase deficiency was detected by inability of blood cells haemolysates to form phosphorylated derivates from galactose-l-14C and to oxidize this substance to14CO2. The activity of galaktose-l-phosphate-urydiltransferase was in normal range. There was a family history of cataracts and the patient showed also opacities in consequence of a 5 week exposure to milk. There were no other symptoms f.e. jaundice, hyperaminoaciduria or vomiting, and no weight-loss. Mellituria disappeared upon elimination of all milk from the diet. The parents were related and apparently gypsies.
