Summary
The quantitative determination of the brain specific protein S-100 by means of immunoelectrophoresis is described. Our immunoelectrophoretic assay uses Litex-agarose as supporting medium and rests on the measurement of the amount of protein by determining the amount of precipitate found in an antigen-antibody reaction. Undiluted anti-S-100 immune serum is added to agarose up to 0.4% before coating the supporting slides. S-100 content should be at least 25 ng/10 μl in order to obtain measurable precipitates. Electrophoresis is carried out for 5 hrs with a current of 1 mA/cm at a temperature ranging between 10 and 15°C. Our results show that by means of a simple immunoelectrophoretic method it is possible to quantitate the S-100 protein and that the sensitivity of the method covers the lower nanogram range.
