Zusammenfassung
Durch β-Strahlen bedingte Veränderungen der DNS-Synthese (autoradiographischer Nachweis durch 3-H-Thymidinmarkierung) und der Mitosetätigkeit des Rattencorneaepithels werden 1/2, 2, 6, 12 und 18 Std nach Applikation von 600, 2400 bzw. 6000 rep untersucht.
Bis zu 6 Stunden nach Bestrahlung ist gegenüber den Kontrollen keine signifikante Änderung des Markierungsindex bei allen Gruppen festzustellen. Nach 12 Std hat sich dieser auf etwa 50% der Kontrollwerte gesenkt und ändert sich bis zum Ablauf der Beobachtungszeit nicht mehr. Diese Veränderungen beruhen wahrscheinlich auf einer Schädigung der Synthese der für die DNS-Bildung notwendigen Enzyme.
Bei der Mitosetätigkeit ist in allen Bestrahlungsgruppen ein sofort einsetzendes, fast völliges Erliegen festzustellen. Ihre Erholung erfolgt dosisabhängig bei den niedrigen Strahlendosen; nach 6000 rep bleibt sie jedoch während der ganzen Beobachtungszeit aus. Als Ursache wird eine Schädigung des Spindelmechanismus diskutiert.
Summary
Changes in the activity of DNA-synthesis (as revealed by radio-autography with 3-H-thymidine) and mitosis are investigated 1/2, 2, 6, 12 and 18 hours after β-ray treatment (600, 2400 and 6000 rep) of the epithelium of the rat's cornea.
Not before 6 hours after irradiation a depression of about 50% is found of the labeling-index. These changes seem to derive from a damage of the synthesis of those enzymes which are necessary for DNA-synthesis.
The mitotic-index is depressed at soon. Its recreation shows a definite relation to the dose applicated. A damage of the cleavage spindle mechanism is discussed.
