Zusammenfassung
In biochemischen und biologischen Labors stellt sich häufig die Aufgabe, den Proteingehalt von wässrigen Lösungen zu ermitteln. Man muss daher die Prinzipien, vor allem aber auch die Vor- und Nachteile der wichtigsten Methoden kennen und sich über ihre – methodisch bedingten – Ungenauigkeit und Fehlerquellen im Klaren sein. Objektive Methoden zur Quantifizierung von Proteinen sind die quantitative Aminosäureanalyse oder die Gewichtsbestimmung des reinen Proteins als Feststoff. Letzteres ist nach Probenaufarbeitung durch Vakuumgefriertrocknung oder durch Hitzetrocknung bei 104–106 °C für 4–6 h möglich. Beide Methoden sind jedoch für die routinemäßige Ermittlung des Proteingehalts einer Lösung zu zeit- und arbeitsaufwendig. Für viele Fragestellungen ist es auch völlig ausreichend, den ungefähren Gehalt, verglichen mit einem definierten Standard wie beispielsweise Serumalbumin, zu kennen. Dies gilt z. B. für Proteinbestimmungen bei der Proteinaufreinigung oder bei der Immobilisierung von Proteinen.
