Zusammenfassung
Die Massenspektrometrie (MS) stellt eine Analysetechnik zur Bestimmung des Masse-zu-Ladungsverhältnisses (m/z) von Ionen im Hochvakuum dar. Ende der Achtzigerjahre des letzten Jahrhunderts führten die Arbeiten von John B. Fenn (USA) zur Entwicklung der Elektrospray-Ionisation (ESI) und von Franz Hillenkamp und Michael Karas (Deutschland) sowie Koichi Tanaka (Japan) zur Entdeckung und Anwendung der matrixassistierten Laserdesorption/Ionisation (MALDI). Erst diese neuen, sanften Ionisationsmethoden machten die intakte Überführung von größeren biologischen Makromolekülen wie Proteinen, komplexen Kohlenhydraten und mehrzähligen Nucleinsäuren in die Gasphase und damit ihre massenspektrometrische Analyse möglich. Im Jahre 2002 wurden Fenn und Tanaka für ihre methodischen Entwicklungen zur Identifizierung von biologischen Makromolekülen mit dem Nobelpreis für Chemie ausgezeichnet. Beide Verfahren sind bezüglich ihrer spezifischen Anforderungen und Stärken komplementär und dominieren zusammen die biologische Massenspektrometrie.
